PIK3CA Mutations and Cancer
The PIK3CA gene encodes the p110a subunit of phosphatidyl 3-kinase, a lipid kinase involved in cell growth, proliferation, motility and survival. It has a central role in the PI3K-AKT-mTOR pathway. Somatic mutations in the PIK3CA gene have been implicated in the pathogenesis of several cancers, including colon cancer, gliomas, gastric cancer, breast cancer, endometrial cancer, and lung cancer.
PIK3CA mutations commonly occur in exon 9, which encodes the helical domain and exon 20, which encodes the kinase domain.

EntroGen’s PIK3CA mutation analysis kit is a polymerase chain reaction (PCR)-based assay that uses allele-specific primers to identify the presence of the PIK3CA somatic mutations. The testing procedure involves three (3) simple steps:
- Isolation of DNA from tumor biopsies, paraffin-embedded sections (FFPE), or fresh frozen tumors
- Amplification of regions of the PIK3CA gene using allele-specific primers
- Detection of amplification product on a Real-Time PCR instrument.
This test can be completed in approximately 2 hours from isolation of DNA to test result.
Equipment and Materials
EntroGen’s PIK3CA mutation analysis kit requires a real-time PCR instrument capable of detecting FAM and VIC fluorescent probes.
This test includes reagents required for the PCR amplification/detection, as well as validated reaction controls. Columns and reagents for DNA isolation are not included.
